Bet365 is currently the largest online bookie in the UK and deservedly so.  They have a simple to use online interface, an easy sign up process and currently offer a deposit match of 200%.

Because other players are laying odds, they tend to be better, especially the longer they are.  A horse that pays out £100 on a £10 bet at a normal bookie, would probably pay out £150 on a £10 bet on a betting exchange.
